Inhibitory activity of dome formation in LLC-PK1 cells is a selective index of aminoglycoside nephrotoxicity.

چکیده
Aminoglycoside antibiotics are widely used clinically for a treatment of Gram-negative bacterial infection by the inhibition of bacterial protein synthesis. However, the aminoglycosides have clinical side effects such as nephrotoxicity and ototoxicity for a long-term treatment. Here we evaluated a simple and useful method for screening a new aminoglycoside without or less nephrotoxicity using LLC-PK1 cells derived from a pig kidney proximal tubular epithelial cell. Dome formation is a morphological trait of LLC-PK1 cells through unidirectional water and salt transport in normal culture condition1,2). In the course of searching for a practical method for evaluating aminoglycosides nephrotoxicity, we reconfirmed usefulness of dome formation in LLC-PK1 cells, which was originally reported by INUI et al.1,2). In this study, we modified and qualified the method for screening a new aminoglycosides, and determined whether it is applied to other antibiotics. As a result, an optimal condition was determined as follows. LLC-PK1 cells obtained from American Type Culture Collection were maintained in a Dulbecco's modified Eagle's medium (DMEM; Nissui Pharmaceutical Co., Ltd.) supplemented with 10% fetal bovine serum

Gentamicin Induced Apoptosis of Renal Tubular Epithelial (LLC-PK1) Cells
Nephrotoxicity is a major limiting factor in the use of aminoglycoside antibiotics, the mechanisms for which are still speculative. To clarify the mechanisms of renal tubular cell death induced by aminoglycosides, we examined the renal proximal tubule-like cell line, LLC-PK1, after inducing apoptosis through a chronic treatment with gentamicin (GM). Changes in the expression of the Fas were als...
متن کاملBeneficial effects of fermented black ginseng and its ginsenoside 20(S)-Rg3 against cisplatin-induced nephrotoxicity in LLC-PK1 cells
BACKGROUND Nephrotoxicity is a common side effect of medications. Panax ginseng is one of the best-known herbal medicines, and its individual constituents enhance renal function. Identification of its efficacy and mechanisms of action against drug-induced nephrotoxicity, as well as the specific constituents mediating this effect, have recently emerged as an interesting research area focusing on...
متن کاملProtective effect of Korean Red Ginseng against FK506-induced damage in LLC-PK1 cells
BACKGROUND Compound FK506 is an immunosuppressant agent that is frequently used to prevent rejection of solid organs upon transplant. However, nephrotoxicity due to apoptosis and inflammatory response mediated by FK506 limit its usefulness. In this study, the protective effect of Korean Red Ginseng (KRG) against FK506-induced damage in LLC-PK1 pig kidney epithelial cells was investigated. MET...
متن کاملProtective Effects of the Fermented Laminaria japonica Extract on Oxidative Damage in LLC-PK1 Cells
This study investigated the protective effect of the butanol (BuOH) fraction from fermented Laminaria japonica extract (BFLJ) on AAPH-induced oxidative stress in porcine kidney epithelial cells (LLC-PK1 cells). L. japonica was fermented by Aspergillus oryzae at 35±1°C for 72 h. Freeze-dried fermented L. japonica was extracted with distilled water, and the extracted solution was mixed with ethan...
متن کاملCisplatin induces apoptosis in LLC-PK1 cells via activation of mitochondrial pathways.
Cisplatin, a commonly used chemotherapeutic agent, has a major limitation because of its nephrotoxicity. Recent studies have shown that cisplatin causes apoptotic cell death in renal tubule cells, but the underlying molecular mechanisms remain to be elucidated. In this study, cisplatin was found to induce apoptosis in a dose- and duration-dependent manner in cultured proximal tubule (LLC-PK1) c...
